变形链球菌亚单位疫苗SBR制备及经颌下腺免疫小鼠的抗体应答 被引量:1

摘要 目的:探讨变形链球菌(S.mutans)唾液结合区段(SBR)作为亚单位防龋疫苗的免疫原性,并评价经涎腺接种诱导有效免疫应答的可行性。方法:构建含S.mutansSBR基因的重组原核表达质粒pTriEx-4-SBR,用该质粒转化大肠杆菌E.coliJM109DE3,培养转化后的E.coli并诱导其表达SBR蛋白;以SBR蛋白作为亚单位防龋疫苗经颌下腺免疫BALBC小鼠,ELISA检测特异性抗体指标。结果:纯化得到的SBR蛋白与预期相符;ELISA结果表明,SBR蛋白经颌下腺免疫小鼠能够诱导血清抗SBR特异IgG(P<0.01)、唾液抗SBR特异IgGP<0.01、唾液抗SBR特异IgAP<0.01显著应答。结论:本实验所表达及纯化的SBR蛋白经涎腺接种途径可诱导明显的免疫应答。 Objective: To confirm the immunogencity of Sreptococcus mutans (S.mutans) SBR as subunit vaccine and to evaluate its feasibility to elicit immune response through vaccination at submandibular gland. Methods: Recombinant prokaryotic expression plasmid pTriEx-SBR containing S. mutans SBR was constructed, and E.coli JM109(DE3) was transformed with the plasmid, and then the transformed E.coli was cultured to express SBR, and fimally SBR protein was purified. Purified SBR was used to immunize BALB/C mice through submandibular gland, and the specific antibody responses were evaluated by ELISA. Results: ELISA showed that immunization with SBR not only can elicit serum IgG, but also specific salivary IgA and IgG. Gonclusion: S.mutans SBR prepared by this study is effective in eliciting immune rseponses through vaccination at salivary gland.
