摘要
目的:探讨卵巢癌细胞SKOV3侵袭转移过程中KAI1基因的表达及意义。方法:采用MTT法观察丁酸钠对细胞的抑制作用,形态学观察SKOV3药物作用后细胞超微结构的变化,及转移抑制基因KAI1的表达情况。结果:药物对卵巢癌SKOV3细胞的抑制率呈剂量依赖性,丁酸钠作用下细胞微绒毛减少变短、消失,KAI1基因免疫组化染色较对照组明显增强。结论:转移抑制基因KAI1在卵巢肿瘤侵袭转移中起着重要作用。
Objective: To study the significence of KAI1 in the invasion and metastesis of human ovarian cancer cell line SKOV3. Mathods: The inhibiting effect of sodium butyrate on cell line was assayed by MTT colormetric assay, the morphologic changes observed under electron microscopes,and variation of KAI1 gene expression investvgated by immunhistochemistry. Results: The inhibiting rate of cell depended on amount of sodium butyrate (NaB). The surface villi lessen shortened and disappeared. Expression of KAI1 gene was significantly stronger than control. Conclution: Sodium butyrate can change the malignant characteristics of ovarian cancer cell line. The kAI1 gene play an inportant role in the metastasis of ovanas cancer.
