摘要
目的 探讨大剂量过敏原对哮喘小鼠T细胞产生IL 5的影响及其机制。方法 应用不同浓度过敏原体外处理哮喘模型小鼠T细胞 ,用单核细胞作抗原递呈细胞。ELISA法检测培养清中IL 5水平 ,并用TUNEL法检测T细胞的凋亡率。结果 ①在 0~ 10 0 μg/ml卵蛋白 (OVA)处理浓度级之间 ,哮喘组小鼠淋巴细胞产生的IL 5的水平随着处理浓度的增高而增高 ,但经高浓度OVA( 10mg/ml)处理后 ,T淋巴细胞产生的IL 5水平却较哮喘低浓度组及对照组明显降低 (P <0 .0 1) ;对照组各浓度级之间则无显著差异 ;②不同浓度OVA处理后哮喘组T细胞的凋亡率均无明显差异。结论 大剂量过敏原可以诱导哮喘T细胞IL 5产生减少为表现的免疫耐受现象 ,其机制主要是诱导淋巴细胞无不反应性 ,而不是诱导T细胞凋亡。
Objective To investigate the effects of allergen at different concentrations on the production of IL 5 by lymphocytes of mice with asthma and its mechanism. Methods The levels of IL 5 produced by T cells treated with allergen of different concentrations and monocytes as antigen presenting cells in vitro were measured by ELISA and the apoptosis rate of T lymphocytes was determined by TUNEL. Results ① The administration of high dose of allergen inhibited the expression of IL 5( P <0.01); ② There was no significant difference in apoptosis rate of T lymphocytes of mice with asthma after treatment with different concentrations of allergen. It was the same case in the control group. Conclusion High dose of allergen can down regulate the production of IL 5 mainly by means of induction of anergy instead of apoptosis of lymphocytes.
