用PKC抑制剂Rottlerin治疗小鼠实验性哮喘的研究 被引量:1

摘要 目的 研究蛋白激酶PKC抑制剂Rottlerin在防治小鼠实验性哮喘中的作用。方法 OVA蛋白诱导BALB/c小鼠建立实验性气道哮喘模型。腹腔注射给予PKC抑制剂Rottlerin与PBS ,比较肺抗性及气道反应 ,肺组织学和Th2 细胞因子产生的差异。结果 腹腔注射Rottlerin ( 0 .3mg/kg)可抑制肺支气管AHR及嗜酸性粒细胞浸润 ,阻止气道组织嗜酸粒细胞炎症 ,Goblet细胞增生及粘液产生 ,明显减弱血清中IgE及Th2 细胞因子的产生。结论 腹腔注射PKC抑制剂Rott Objective To study the potential role of PKC inhibitor Rottlerin in the treatment of asthma. Methods Balb/c mice were sensitized and challenged with ovalbumin(OVA) protein to construct murine experimental model of asthma. Then the mice were treated by means of injection of Rottlerin or phosphate buffered saline (PBS) into the abdominal cavity. Changes of the total serum IgE, pulmonary eosinophils, cytokines and pulmonary inflammation were investigated. Results Injection of Rottlerin at the dose 0.3 mg/kg into the murine abdominal cavity could inhibit the infiltration of pulmonary eosinophils and pulmonary inflammation and significantly decrease the production of total serum IgE and Th 2 cytokines as well. Conclusion Rottlerin can inhibit murine experimental asthma.
