摘要 目的 :探讨MAPK在慢性髓细胞白血病 (CML)信号转导中的作用及其作用机制 .方法 :用脂质体转染法将MAPKASO导入K5 6 2细胞中 ,通过集落形成、DNA合成、蛋白质含量和MAPK活性的变化检测MAPKASO对K5 6 2细胞的作用 .结果 :MAPKASO可明显抑制细胞集落形成、DNA合成、蛋白质含量和MAPK活性 ,其抑制率分别为 4 6 .1% ,5 7.1% ,4 3.2 %和 6 2 .0 % ,与对照组比较有显著差异 (P <0 .0 5 ) .结论 :MAPK在CML信号转导中起重要作用 。 AIM: To study the effects and mechanism of mitogen activated protein kinase (MAPK) in cell signal transduction of chronic myelogenous leukemia (CML). METHODS: After MAPK antisense oligodeoxynucleotide (ASO) was introduced into K562 cell line by liposomal transfection, the effects were evaluated by colony formation, DNA synthesis, protein content and MAPK activity. RESULTS: The colony formation, DNA synthesis, protein content and MAPK activity were significantly inhibited by MAPK ASO, and the inhibitory rate was 46.1%, 57.1%, 43.2% and 62.0% respectively ( P <0.05). CONCLUSION: MAPK plays an important role in the signal transduction of CML and MAPK can be used as a new target in the treatment of CML.
