垂体瘤转化基因的表达及其与肿瘤侵袭性的关系 被引量:11

摘要 目的 拟对垂体瘤转化基因 (pituitarytumor transforminggenePTTG)及与之密切相关的碱性成纤维生长因子 (bFGF)在垂体瘤中的表达进行研究 ,并探讨其可能的临床意义。方法 取垂体腺瘤组织 30例 ,4例正常垂体。从组织中提取总RNA ,经RT PCR方法扩增PTTGmRNA ,采用免疫组化方法测定bFGF、增殖细胞核抗原 (PCNA)及微血管密度 (MVD) ,分析各指标和临床特征的关系。结果 侵袭和非侵袭组的PTTGmRNA、PCNA、bFGF及MVD相比均有显著差异性。bFGF和MVD、PC NA、PTTGmRNA之间存在显著正相关。结论 PTTG参与垂体瘤的早期发生。bFGF则对血管形成 ,肿瘤增殖有明显作用 ,二者均和肿瘤的侵袭性密切相关。PTTG→bFGF→肿瘤增殖可能为垂体瘤发生、发展的重要机理。 Objective To study PTTG and bFGF expressino in pituitary adenomas, and investigate the differences between invasive group and non-invasive group. Methods Thirty pituitary tumor specimens were obtained at surgery and frozen in -70℃ refrigenrator.Total RNA was extraced from specimens and PTTGmRNA was detected by RT-PCR.bFGF,PCNA and CD34 were studied by immunocytochemical method. Results PTTGmRNA,bFGF,PCNA and MVD were significantly different between invasive group and non-invasive group. bFGF and MVD,bFGF and PCNA ,PTTGmRNA and bFGF have positive correlation (P<0.01)Conclusions PTTG may be involved in the early stage of pituitary tumorigenesis and bFGF was important factor in tumor angiogenesis and proliferation. PTTG→bFGF→turmor proliferation may be importand mechanism in pituitary adenomas.
