摘要
目的 :观察NIH3T3细胞中BMP信号转导因子的表达 .方法 :用免疫组化和原位杂交方法检测NIH3T3细胞中BMP ,BMPⅠ ,Ⅱ型受体和Smad1,4 ,5的表达 ,在细胞培养液中加入 5 0 μg·L-1rhBMP 2 ,检测细胞的碱性磷酸酶 (ALP)活性和骨钙素 (OC)含量观察细胞是否有成骨趋向 .结果 :细胞中有BMPⅠ ,Ⅱ型受体和Smad1,4 ,5的表达 ;没有BMP的表达 ;加入 5 0 μg·L-1rhBMP 2后 ,ALP活性和OC含量均增高 .结论 :rhBMP 2作用于NIH3T3细胞后 ,细胞有成骨趋向 .NIH3T3细胞是rhBMP 2的靶细胞 。
AIM: To examine the BMP 2 induced signal transduction in NIH3T3 cells. METHODS: The expressions of BMP, BMPRⅠ, BMPRⅡand Smad1, 4, 5 were examined in NIH3T3 cells by immunohistochemistry and in situ hybridization. The NIH3T3 cells were cultured in DMEM with 50 μg·L -1 rhBMP 2 and the ALP activity and osteocalcin (OC) quantity of NIH3T3 cells were investigated. RESULTS : The NIH3T3 cells showed positive signals of BMPRⅠ, BMPRⅡand Smad1, 4, 5 and negative BMP signal. The NIH3T3 cells cultured with 50 μg·L -1 rhBMP 2 showed enhanced ALP activity and OC quantity. CONCLUSION: 50 μg·L -1 rhBMP 2 can transform NIH3T3 fibroblasts to osteo like cells and BMP 2 induced signal transduction system exists in NIH3T3 fibroblasts.
