机械压力对兔髁突软骨碱性磷酸酶活性及细胞增殖的影响 被引量:6

摘要 目的 :探讨不同作用时间及不同液压力值对髁突软骨细胞碱性磷酸酶活性及细胞增殖的影响 .方法 :消化法培养 2wk新西兰白兔的髁突软骨细胞 ,用自行设计制作的可控液压细胞加载装置在 30 ,6 0 ,90kPa压强下加载 6 0 ,36 0 ,72 0min ,用酶动力学方法检测细胞ALP活性 ,MTT法检测细胞增殖水平 .结果 :正常髁突软骨细胞随体外培养时间的延长 ,ALP活性持续增加 .30kPa组ALP活性与相应时间点的对照组相比无显著差别 ,但在加压 72 0min后细胞增殖显著受到抑制 ;6 0kPa组在加压至 36 0min时ALP活性较对照组显著升高 ,同时细胞增殖速率显著降低 ;90kPa组从加压 6 0min起ALP活性就有显著升高 ,但当加力时间延长至 72 0min时 ,ALP活性反而稍有下降 ,但仍明显高于对照组 ,该组持续受压 36 0min及 72 0min细胞的细胞增殖速率均较对照组显著降低 .结论 :6 0kPa持续加压 36 0min或 90kPa持续加压 6 0min以上可使髁突软骨细胞的ALP活性显著升高 。 AIM: To investigate the effects of mechanical pressure on the activity of alkaline phosphatase and function of mandibular condylar chondrocytes (MCC). METHODS: In vitro cultured MCC from two week old New Zealand rabbits were incubated under continuous pressure of 30, 60 and 90 kPa for 60, 360 and 720 min. The activity of alkaline phosphatase (ALPase) was determined by modified enzyme dynamical method and the cell proliferation was detected by MTT assay. RESULTS: Under the pressure of 60 kPa for 720 min, the ALPase increased obviously while the function of cells in that group decreased. In the group of 90 kPa, the ALPase increased more significantly than that of the control under the pressure for 360 min, and it decreased slightly when the pressure continued for 720 min, but still higher than that of the control. The A value of cells under the pressure of 60 kPa for 720 min or 90 kPa for more than 360 min was lower than that of the control. CONCLUSION: Continuous pressure of 60 kPa for 720 min or 90 kPa for more than 360 min can stimulate the activity of ALPase and inhibit the proliferation of MCC.
