摘要
目的 :研究大鼠小肠缺血再灌注后肝组织中Bax,Bcl 2 ,p5 3的表达及超微结构变化 ,探讨小肠缺血再灌注后肝组织的损伤 .方法 :建立小肠缺血再灌注模型 ,分对照组 ,再灌注后 0 ,30min ,1,2h ,1,3,7d共 8组 ,用免疫组织化学SP法观察肝组织中Bax ,Bcl 2和p5 3的表达情况 ,透射电镜观察肝细胞超微结构变化 .结果 :Bax,Bcl 2 ,p5 3阳性细胞主要在肝血管窦内皮细胞和肝细胞中表达 .再灌注 30min ,Bax ,Bcl 2 ,p5 3阳性细胞率均明显升高 ,分别为 11.3% ,2 4 .8% ,7.1% ,Bcl 2阳性细胞率明显高于Bax阳性细胞率 ,两者差别显著 (P <0 .0 1) .2h时降低 ,其后持续升高 ,7d时阳性细胞率达到最高水平 ,Bax ,Bcl 2 ,p5 3阳性细胞率分别为6 3.3% ,4 1.3% ,38.5 % ,Bax阳性细胞率明显高于Bcl 2 ,两者差别显著 (P <0 .0 1) ,透射电镜显示肝细胞超微结构损伤改变 .结论 :Bax ,Bcl 2 ,p5
AIM: To study the expression of Bax, Bcl 2, p53 and the changes of ultrastructure in the liver and to find out the injuries of the liver after small intestine ischemia reperfusion in rats. METHODS: Models of ischemia reperfusion of small intestine were established at 0, 30 min, 1, 2 h, 1, 3, 7 d after reperfusion. The expression of Bax, Bcl 2 and p53 in the liver was observed by immunohistochemical SP method and the changes of ultrastructure in the liver was observed by transmission electron microscopy. RESULTS : The positive cells were found in sinus endothelial cells and hepatocytes. After 30 min reperfusion, the rate of positive cells of Bax, Bcl 2 and p53 increased to 11.3%, 24.8% and 7.1%, with the rate of positive cells of Bcl 2 higher than that of Bax( P <0.01)but at 2 h, they decreased apparently, then increased and reached the peak of 63.3%, 41.3% and 38.5% respectively 7 d after reperfusion, with the rate of Bax higher than that of Bcl 2( P < 0.01 ). Transmission electron microscopy showed injuries of ultrastructure in the liver. CONCLUSION: The expression of Bax, Bcl 2 and p53 and the ultrastructural changes suggest the apoptosis and injuries in the liver after small intestine ischemia reperfusion in rats.
