钙拮抗剂DCDDP对大鼠肺动脉高压中端粒酶逆转录酶的抑制作用 被引量:4

摘要 目的 :探讨钙拮抗剂 2 ,6 二甲基 4 (2 氯苯基 ) 1,4 二氢 3,5吡啶二羧酸二甲酯 (DCDDP)对大鼠野百合碱(monocrotaline ,MCT)性肺动脉高压 (PH)肺组织中端粒酶逆转录酶 (telomerasereversetranscriptase,TERT)表达的影响 .方法 :用MCT复制大鼠MCT性慢性PH病理模型 ,DCDDP 5 ,5 0和5 0 0 μg·kg-1·d-1ip连续 2 8d ,用免疫组化法和图像分析技术测定肺组织中TERT的表达 .结果 :TERT可在正常组大鼠肺血管平滑肌、支气管平滑肌和软骨组织中表达 ,DCDDP组比MCT组肺组织中TERT表达降低 ,DCDDP低剂量组 (5 μg·kg-1·d-1)和中剂量组 (5 0 μg·kg-1·d-1)尤为明显 .结论 AIM: To explore changes of telomerase reverse transcriptase (TERT) on chronic pulmonary hypertension induced by monocrotaline (MCT) in Sprague Dawley rats. METHODS: The rats were treated by dimethyl 4 (2 chlorophenyl) 1,4 dihydro 2,6 dimethyl 3,5 pyridinedicarboxylate (DCDDP) and TERT was detected by immunohistochemistry and image analysis. RESULTS: TERT was positive in pulmonary vascular smooth muscle cells, bronchial smooth muscle cells and chondrocytes in control group. The positive expression of TERT in DCDDP group was less than that in MCT group in pulmonary vascular smooth muscle cells and bronchial smooth muscle cells. CONCLUSION: DCDDP inhibits the over expression of TERT of the chronic pulmonary hypertension induced by MCT in rats.
