一种基于电化学发光技术的高灵敏度PS-1基因点突变检测方法 被引量:5

摘要 电化学发光(electrochemiluminescence,ECL)分析是20世纪90年代初发展起来的一种高灵敏度检测方法,它将电化学法和化学发光法巧妙结合,利用金属钌的复合物和三丙胺在电极表面发生氧化还原反应而产生高效、连续、稳定的发光,具有灵敏、快速、准确、操作简便和分析适应性广等特点,已被应用于核酸的定量分析之中。将电化学发光技术应用于基因点突变检测,采用了一种基于PCR技术和限制性内切酶技术的电化学发光分析方法检测Presenilin-1基因(简称PS-1基因)密码子235处发生的点突变。结果显示,在PCR循环次数相同的条件下,野生型样品酶切前后的电化学发光信号强度基本保持不变;突变型样品酶切前后的电化学发光信号强度变化显著。该方法可明显区分野生型样品和突变型样品,可望用于老年性痴呆病的早期诊断。
