摘要
为探讨 17β 雌二醇 ( 17β estrodiol,17β E2 )及植物雌激素α 玉米赤霉醇 (α zearalanol,α ZAL)对人脐静脉内皮细胞 (Humanumbilicalveinendothelialcells ,HUVECs)组织因子 (tissuefactor ,TF)蛋白含量及mRNA表达的影响 ,进行人脐静脉内皮细胞原代培养 ,传至 2~ 3代后用于实验。在细胞培养液中分别加入不同浓度 17β E2 及α ZAL作用 48h ,采用酶联免疫吸附法 (ELISA)测定细胞裂解液中TF蛋白浓度 ,双辛丁酸法 (bicinchoninicacid ,BCA)检测细胞总蛋白含量。提取细胞总RNA ,用RT PCR观察 17β E2 及α ZAL对TFmRNA表达的影响。结果发现 :10 -7mol L 17β E2 及α ZAL均可明显降低HUVECs组织因子蛋白含量及mRNA的表达 ,二者的作用类似。表明 17β E2 及α ZAL可明显抑制HUVECs组织因子的表达 ,这可能是它们抗血栓形成。
To investigate the effects of 17β-estradiol and phytoestrogen α-zearalanol on tissue factor(TF) protein levels and mRNA expression in cultured human umbilical vein endothelial cells (HUVECs). HUVECs were treated with 17β-estradiol and α-zearalanol for 48 hours. ELISA and RT-PCR were applied to study the changes of TF protein and mRNA respectively. We found that both 10 -7mol/L 17β-estradiol and α-zearalanol markedly decreased tissue factor protein levels and mRNA expression. 10 -6mol/L angiotensin Ⅱ induced TF protein levels and mRNA expression apparently, while co-incubation with 17β-estradiol and α-zearalanol might reverse this change. These results suggest that both 17β-estradiol and phytoestrogen α-zearalanol can inhibit tissue factor expression in HUVECs, and there is no significant difference in their effects.
