新生大鼠脊髓损伤后再生相关基因文库构建及EST筛选 被引量:2

摘要 目的:分析新生大鼠损伤后再生时运动皮层神经元基因表达变化以探讨相关的再生机制。方法:新生大鼠脊髓双侧半横断后即刻单侧胚胎脊髓移植,术后3天用抑制性消减杂交技术筛选大鼠双侧运动皮层神经元差异表达基因,构建差减cDNA文库,通过蓝白斑实验和cDNA斑点杂交验证差异表达基因真阳性克隆,测序分析。结果:发现25个全新的EST片段,在GenBank中登陆并获得注册。结论:新生大鼠皮层神经元轴突中断后的再生过程中有基因表达变化,变化的基因可能与神经元再生有关。 Objective: To identify the differential expression genes of the regenerating somatosensory cortex after the spinal cord of the newborn rats was hemi-sectioned, to clone them and analysis their relationship with regeneration.Method: The regeneration model of the newborn rats whose bilateral spinal cord were hemi-sectioned was used. The fetal spinal cord tissue was transplanted at their unilateral lesion site. The suppression subtractive hybridization method was used to screen the differential expression genes of motor cortex neuron of the newborn rats. The differential expression library was constracted. The blue/white spot test and dot hybridization technique was used to certificate the really positive clones. Then their sequences was analysed. Result: 25 novel ESTs were discovered and got accession in GenBank.Conclusion:The somatosensory cortex neuron of the newborn rats with transplanted fetal spinal cord have a differential expression of the regeneration relation genes, the differential genes may influence the regeneration reaction.Author′s address Research Institute of Surgery, Third Military Medical University, Chongqing 400042
