摘要
从 Gen Bank数据库下载了世界各地分离的猪圆环病毒基因组全序列 ,并对其进行多重比对 ,绘制系统进化树 ,发现来自同一国家或地区的 PCV毒株亲缘关系较近。提取 PCV ORF1编码的蛋白质序列 ,进行多序列比较 ,分析保守氨基酸序列 ,搜索结构域相似序列 ,结果发现玉米线条病毒属与 PCV的起源有密切的关系。此外 ,在数据处理过程中通过自编程序结合互联网及免费软件 ,能迅速进行大规模序列比较分析 (35个序列以上 ) ,提高了生物数据分析处理的速度。
Fifty-one genomic sequences of porcine circovirus(PCV) from different countries all over the world were downloaded from GenBank and transformed to standard sequence format.Multiple alignment indicated that the PCV-2 strains from same district or a country clustered in the same or neighboring branches in the phylogenetic tree.The amino acid sequences of ORF1 protein were further extracted from these sequences and aligned.The consensus sequence was submitted to SCANPS WWW server to search the structure counterparts.The results showed that Mastrevirus have most probability relationship with PCV′s evolutional origin.In addition,during the procedure of large scale sequence comparison analysis,the speed of research had been improved by programming scripts together with internet and free software.
