摘要 目的:介绍基因转移载体的研究进展,为其深入研究提供参考。方法:对近年来有代表性的文献进行分析、归纳。结果:介绍了病毒载体和非病毒载体的主要优缺点、研究方向及存在的障碍。结论:病毒载体虽转染率较高、表达时间长,但其安全性令人担忧。非病毒载体的转染率还不够高,有效表达时间短,其毒副反应近来也受到关注;为了进一步提高其转染率和表达持续时间,还须深入研究外源DNA进入靶细胞核的细胞和分子机制。 Objective:To introduce the progress of gene transfer vectors for its further study.Methods:The typical articles in recent years have been reviewed and classified.Results:The advantage and disadvantage of viral and non- viral vectors have been highlighted. The viral vectors have higher transfer efficiency and longer duration of gene expression than non- viral vectors,but its safety issues surrounding the use of viruses have been plagued. Conclusion:It is necessary that the cellular and molecular mechanism of delivering DNA to the nuclear of targeting cell be intensively studied in order to improve the transfer efficiency and the gene expression duration of non- viral vectors.
