肾络通对肾小管间质纤维化大鼠肾素血管紧张素系统的影响 被引量:15

The Effect of Shenluotong (肾络通) on rennin-angiotensin system in adriamycin induced renal tubule-interstitial fibrosis rats

摘要 目的 :观察肾络通对阿霉素诱导的肾小管间质纤维化大鼠肾组织肾素血管紧张素系统 (RAS)及转化生长因子 β1(TGF β1)、TGF β1mRNA表达和纤维连接蛋白 (FN)、层粘连蛋白 (LN)的影响。 方法 :采用生化、放射免疫法及免疫组织化学、原位杂交等方法观察大鼠肾功能 ,肾组织肾素活性、血管紧张素Ⅱ(ANGⅡ)水平及TGF β1、FN、LN的蛋白表达和TGF β1mRNA表达。 结果 :肾络通及缬沙坦组肾功能明显改善 ,肾素活性、ANGⅡ水平有所下降 ,TGF β1、FN、LN的蛋白表达及TGF β1mRNA减少。 结论 :肾络通可能通过调节RAS ,抑制TGF β1和TGF β1mRNA的表达 ,减少细胞外基质的沉积 ,减轻肾小管间质纤维化进程。 Objective: To observe the effect of Shenluotong on rennin-angiotensin system(RAS), and the expression of TGF-β1, TGF-β1mRNA, LN and FN in adriamycin induced renal interstitial fibrosis model rats. Method: Rats were treated by Shenluotong and varsartan for 10 weeks. BUN ,Scr, 24 hours proteinuria, activity of rennin and angiotensin II, TGF-β1, FN and LN were measured by radio-immunity , immunohistochemical staining and in situ hybridization. Result: Scr, BUN in serum, 24 hours protein andβ2-MG in urine were reduced markedly by Shenluotong and valsartan compared with model group, expressions of TGF-β1, LN and FN in protein level and TGF-β1 in mRNA level were down-regulated in treating groups. Conclusion: Shenluotong has the effect to inhibit the adriamycin induced renal interstitial fibrosis rats by regulated activity of rennin-angiotensin system ,down-regulated the expression of TGF-β1 in protein and mRNA levels and reduce the deposition of extracellular matrix.
