摘要
在确立葡萄卷叶病毒Ⅲ和扇叶病毒RT-PCR优化体系基础上,研究了多重PCR的主要成分:dNTPS、TaqDNA聚合酶、MgCl2对PCR反应的影响,从而确立了GLRaVⅢ、GFLV的多重RT-PCR检测体系。研究结果表明,多重PCR反应体系中TaqDNA聚合酶的用量较单重PCR反应体系要低,而且可降低成本和缩短检测时间。
Based upon the establishment of a optimized RT-PCR detection system of GLRaVⅢand GFLV,the reaction of the main ingredients of multiple PCR:dNTPs ,Taq E,prime ,Mg 2+ was examined and optimized,and so established the mul-tiple PCR system.Th is system can detect2viruses simultaneity.Studies indicated that the Taq E use d in multiple PCR could be less than the optimum amount ,so it is economical an d timesaving.
