摘要
目的:研究不同培养基中兔关节软骨细胞生物学行为的差异,为软骨组织工程中软骨细胞的培养寻找最合适的培养基。方法:取3个月龄新西兰兔关节软骨消化获取软骨细胞,以相同密度分别用两种培养基(F-12、1640)进行培养,观察其形态、生物学行为、增生速度、特征性产物分泌量及黏附能力。分析两种培养基培养结果,对比其对体外培养关节软骨细胞的影响。结果:软骨细胞在1640培养基中贴壁、生长较快,细胞形态均一,特征性产物分泌旺盛。结论:在两种培养基中1640培养基更适合进行关节软骨细胞体外培养。
Objective:To study the biological behaviors of rabbit articular c ho ndrocytes cultured in different culture mediums,and provide the most ideal cult ure medium for cartilage tissue-engineering. Methods: Chondrocytes from the joi nts of 3-month-old New Zealand rabbits were harvested,growing in F-12 and 16 40 mediums separately with just the same cell density. Then,the morphology, bio logical behaviors, proliferation speed, martrix synthsis, and the ability of att achment of chondrocytes in different culture mediums were observed. Finally,the effect of F-12,1640 culture medium on normal rabbit articular chondrocytes cul tured in vitro were compared. Results:The proliferation speed, morphology,martri x synthsis and attachment of rabbit articular chondrocytes in 1640 medium were b etter than in F-12 medium. Conclusion: The culture medium 1640 is more suitable for rabbit articular chondrocyte culture in vitro. [
