摘要
目的 探讨硫代乙酰胺诱导的肝硬化大鼠GH/IGF - 1轴的变化。方法 设正常组大鼠 6只、肝硬化组大鼠 6只 ,测大鼠肝功能、血糖和血清GH、IGF - 1、IGFBP - 3水平 ,用RT -PCR法检测肝ALBmRNA、IGF- 1mRNA、IGFBP - 3mRNA表达。肝组织行HE染色和Ki6 7免疫组化染色。结果 肝硬化组大鼠血清AST、ALT、ALP明显高于正常组 ,血清TBIL、ALB、血糖两组间无明显差别 ,血清GH水平明显高于正常组 ,而血清IGF - 1、IGFBP - 3水平明显低于正常组 ,肝ALBmRNA、IGF - 1mRNA、IGFBP - 3mRNA表达水平明显低于正常组。肝硬化组大鼠镜下见假小叶形成 ,肝Ki6 7指数明显高于正常组。结论 肝硬化大鼠存在GH/IGF -
Objective:To explore the changes in growth hormone (GH)/insulin like growth factor-1 (IGF-1) axis of rats with thioacetamide-induced liver cirrhosis. Methods:Six rats with thioacetamide-induced liver cirrhosis served as cirrhotic group and six normal rats as normal group. Liver function,blood glucose, serum GH, IGF-1 and IGF binding protein-3(IGFBP-3) were detected. Albumin (ALB) mRNA, IGF-1 mRNA and IGFBP-3 mRNA levels in liver were detected by RT-PCR. Liver pathological observation included HE staining and Ki67 immunohistochemistry staining.Results:In the cirrhotic group, serum AST, ALT, ALP GH levels were significantly higher but serum IGF-1 and IGFBP-3 levels were significantly lower and the expressions of hepatic ALB mRNA, IGF-1 mRNA and IGFBP-3 mRNA were significantly lower than those in normal group. Liver Ki67 labeling index was significantly higher in cirrhotic group.Conclusions:There is an obvious change in GH/IGF-1 axis of rats with thioacetamide-induced liver cirrhosis. Dectecting the change of GH/IGF-1 was helpful to choose nutrients.
