大鼠神经干细胞移植治疗大鼠脑梗死的实验研究 被引量:20

摘要 目的 研究大鼠神经干细胞治疗脑梗死的可能性。方法 从孕 16d的大鼠胚胎脑组织中分离、培养神经干细胞 ,通过免疫组织化学技术研究其特征。制作大鼠脑缺血再灌注模型 ,3d后移植未分化的神经干细胞于梗死灶同侧的侧脑室内。记录损伤和移植后的大鼠运动功能 ,不同时间杀死大鼠 ,研究移植后的神经干细胞迁徙、分化的情况。结果 大鼠神经干细胞体外培养显示其具有良好的自我繁殖能力 ,体外诱导可分化为神经元细胞。结论 大鼠神经干细胞能在缺血区内分化成为神经元细胞 ,移植后能有效穿透室管膜进入脑组织 ,迁徒至脑缺血病灶 ,侧脑室移植神经干细胞能有效改善脑缺血动物运动功能。 Objective:To explore the feasibility of the rat neural stem cells transplantation to treat cerebral ischemia.Methods:Neural stems were separated from P16 days brains of rats and were cultured and induced to differentiate.The cerebral ischemic rat models were made and the neural stem cells were transplanted into the ispsilateral ventricles 3 days later.The motor function of rats was investigated.Rats were sacrificed at different time.The differentiation and migration of neural stem cells were checked after transplantation.Results:Neural stem cells showed well ability of multiply in vitro cultivation.These cells could differentiate to be nerve cell through outside the body inducement.Conclusion:The differentiation of neural stem cells in injured area were checked.These cells could pass ipsilateral ventricles effectively,and migrated to the injured area.Nerual stem cells transplantation ipslateral ventricles to the cerebral ischemia could effectively improve the motor function of amimals with cerebral ischemia.
