摘要
目的 探讨对氧磷酯酶 (paraoxonase ,PON) 1基因Q191R多态性与 2型糖尿病合并高血压的关系。方法 以PCR扩增目的片段 ,PCR产物用限制性内切酶AlwⅠ酶切和 2 %琼脂糖电泳分离。分光光度法检测血清PON活性。以放免法测定血清胰岛素和C肽。结果 (1) 2型糖尿病合并高血压组QQ基因型和Q等位基因频率高于单纯 2型糖尿病组 (分别P <0 .0 5和P <0 .0 1) ;(2 )不同等位基因和基因型血清PON活性差异存在显著性 :Q等位基因携带者的血清PON活性低于R等位基因携带者的血清PON活性 (P <0 .0 1) ,PON活性按以下基因型顺序增高QQ <QR <RR(P <0 .0 1) ;(3 )不同疾病状态下血清PON活性差异存在显著性 ,2型糖尿病合并高血压组血清PON活性低于单纯 2型糖尿病组、原发性高血压组及正常对照组 (均P <0 .0 1) ;(4 )Logistic回归分析表明 ,空腹血糖和PON活性是 2型糖尿病合并高血压的独立变异危险因素。结论 PON1基因多态性通过改变血清PON活性而影响糖尿病合并高血压的发生。
Objective To investigate the relation between paraoxonase (PON) 1 Q191R gene polymorphism and type 2 diabetes mellitus complicated with hypertension. Methods Genomic DNA was extracted from peripheral blood leukocytes. The studied DNA was amplified by PCR, and then digested with the restriction endonuclease AlwⅠ and separated on 2% agarose gel electrophoresis. PON activity was estimated by spectrophotometer. Serum insulin and C-peptide were measured by radioimmunoassay. Results The frequencies of QQ genotype and Q allele in type 2 diabetes mellitus with hypertension group were significantly higher than those in type 2 diabetes mellitus group (P<0.05 and P<0.01 respectively). PON activity was associated with PON genotype, serum PON activity increased in the order of QQ<DR<RR (P<0.01). PON activity in type 2 diabetes mellitus with hypertension group was lower than that in type 2 diabetes mellitus group and essential hypertension group and control group (all P<0.01). Logistic regression analysis suggested that fasting plasma Glucose and PON activity were the independentriskfactorsofdiabetesmellitus with hypertension. Conclusion PON1 Q191R gene polymorphism is related to diabetes mellitus with hypertension via influencing the PON activity.
