摘要
从芫荽(Coriandrum Sativum L.)花叶病株上分离到一病毒分离物(CMV-Co),汁液摩擦接种和桃蚜(Myzus Persicea)均能传毒。人工摩擦接种可侵染5科21种植物,在黄瓜、心叶烟等植物上引起系统花叶;在苋色藜、昆诺阿藜、豇豆等接种叶片上引起局部枯斑。此病毒的钝化温度为55~60℃,稀释限点为10^(-3)~10^(-4),体外存活期2~3天。部分提纯的病毒最大紫外吸收为258nm,最小吸收为238nm,表现为典型的核蛋白吸收,A260/A280=1.26。SDS-不连续梯度聚丙烯酰胺凝胶电泳解离病毒其外壳蛋白亚基为一条多肽链,分子量约为25 500道尔顿。琼脂免疫双扩散试验表明,CMV-Co与黄瓜花叶病毒有明显的血清学反应:用1.5%醋酸双氧铀负染,电镜观察病毒粒子球形(正二十面体),直径为28~30nm。根据上述特征,CMV-Co是黄瓜花叶病毒的一个株系。
A virus was isolated from the corider (Corianclrum Sativum L.) with the symptom of mosaic in suburb of Taian. It has been identified as a corider isolate of cucumber mosaic virus (CMV-Co) on the basis of its host rang, transmission, stability, particle morphology, serology, ultraviolet absorption spectrum and molecular weight of coat protein subunit This virus could infect 21 plant species belong to 5 families by sap inoculation. It could be transmitted by aphid (Mazus persicae) in a non-persistant manner. Its DEP is about 10^(-3)~10^(-4), TIP is 55℃~60℃ and L is 2~3 days. the virus particles are isometric with diameter 28~30nm. the ultraviolet absorption spectrum of purified preparation of CMV-Co are typical of nucleoprotein with rations of absorbance at 260nm to that at 280nm of 1.62. One protein band appeared by electrophoresis in SDS-discontinous polyacrylamide gels and estimated their molecular weight to be about 2.55×10^(-4)dalton. in gel double diffusion test, specific precipitation line was formed between the antiserum to CMV and our isolate.
