人参二醇组皂甙对人视网膜色素上皮细胞增生的抑制作用 被引量:6

摘要 目的 :研究人参二醇组皂甙 ( panaxadiol saponins,PDS)对体外培养视网膜色素上皮( RPE)细胞增生的直接抑制作用及可能机制。方法 :通过活细胞计数法与 MTT比色法分别检测不同浓度的 PDS和 2 0 0 mg· L-1PDS在不同作用时间 ( 6~ 1 2 0 h)对 RPE细胞增生及代谢的影响。结果 :PDS组细胞增殖明显受抑并出现细胞脱落 ,40 0 mg· L-1PDS具有最强的抑制效应 ,其明显抑制效应在 6h出现 ,96h达高峰。PDS组 A值明显低于对照组 ,RPE细胞生存率下降。结论 :PDS可能通过阻滞钙通道降低细胞内钙浓度、干扰 RPE细胞代谢 ,对 Objective:To investigate the effects of Panaxadiol saponins (PDS) on proliferation of cultured retinal pigment epithelium cells and the probable mechanism. Methods:Cultured RPE cells were treated with PDS by varied concentrations (500,400,300,200,100,50,10 mg·L -1 ) and with PDS 400 mg·L -1 at various times (6~120 h), cells proliferation and metabolism were evaluated by cell count method and MTT colorimetric assay.Results:PDS showed inhibitory effect on proliferation of RPE cells and some cells detached from the wall in PDS group. The best concentration of PDS was 400 mg·L -1 . The significant inhibitory effect of PDS occurred at 6 h, and the strongest inhibition of PDS occurred at 96 h. The A value and the survival rate of RPE cells in PDS groups was significantly lower than those in control group. Conclusion: The results suggested that PDS could inhibit the proliferation of RPE cells in a concentration dependent and time dependent manner, the probable mechanism was that PDS could block the calcium channel so intracelluar concentration of calcium was lowered and the metabolism of cell was disturbed.
