摘要
对西洋参悬浮培养细胞的生长,各种生长素(2,4-D,NAA、IBA和TAL)中以2,4-D、IBA和TAL较好,二种细胞分裂素(KT、BA)中以BA较好。对西洋参悬浮培养细胞中粗皂甙含量,当IBA浓度在3.5mg/L以下时,粗皂甙含量随IBA浓并的增加而增加,IBA的较适浓度为3.5mg/L_在2,4-D、BA、IBA、TAL、NAA和KT的组合试验中,以2.4-D 0.5、IBA 3.5、BA 0.5(mg/L)的组合对生长最好,其次是KT 0.1、2,4-D 0.5(mg/L)的组合,适量的LH和CM均促进细胞生长。硝态氮有利细胞生长,而铵态氮则有利皂甙的产生。将培养基中蔗糖用量由2.5%0降到1.5%时,人参皂甙产率由39.4mg/L增至138.4mg/L,糖的使用方式对于生长也很重要。
A series of chemical factors affecting the growth and the saponin content of Panax quinquefolium cultured cells in a suspension medium were trialed in this study. The results showed that, for the growth of cells,the effects of some auxins like 2,4-D, IBA and TAL were more efficient than that of NAA. While between two phttokinins, BA was better than KT. As for the total saponin content of cultured cells, it was increased as the increase of IBA with its concentration by 3.5mg/L. in the medium. The optimum concentration of IBA was 3.5mg/L.. In a trial with comopound factors, the cell growth was best in the treatment of 2,4-D 0.5+IBA 3.5+BA 0.5(mg/L.),followed by the treatment of 2,4-D 0.5+KT 0.1(rag/L.). Using appropriate amount of LH and CMcould stimulate the cell growth, As to the nitrogen source NO_3-N was more beneficial for the cell growth than NH_4-N,though the latter was advantageous to the saponin yield. When the surcrose concentration was reduced in the medium from 2.5% to 1.5%, the saponin yield was increased from 39.4mg/L. to 138.4mg/L., for the cell growth,using method of sucrose and glucose was importnt.
