S100A4反义核酸抑制神经母细胞瘤细胞侵袭的作用 被引量:4

摘要 目的 探讨S10 0A4基因在神经母细胞瘤 (NB)细胞侵袭和转移中的作用及其可能机制。方法 将S10 0A4反义核酸转染NB细胞系LA N 6,RT PCR检测S10 0A4、基质金属蛋白酸 (MMP 2 )mRNA水平 ,Transwell检测细胞迁移和侵袭能力。结果 转染反义核酸的LA N 6细胞S10 0A4、MMP 2mRNA表达与对照组相比 ,分别下调 3 5.6%和 2 5.5% ,趋化和侵袭细胞数较对照组明显减少 (P均 <0 .0 5)。结论 反义封闭S10 0A4基因可抑制LA N 6细胞迁移和侵袭力 ,伴MMP 2mRNA表达减低 ,因此 ,S10 0A4基因在NB侵袭转移过程中发挥作用 ;其作用机制可能与增加瘤细胞运动能力 ,调节MMP Objective To investigate the effects and mechanism o f S100A4 gene on the invasion and metastasis of neuroblastoma cells.Meth ods A 20mer phosphorothioate oligodeoxynucleotides targeted against the S100A4 mRNA were transferred into LA-N-6 cells by Lipofectamine 2000. The S10 0A4 mRNA and MMP-2 mRNA expression were detected by reverse transcription polym erase chain reaction(RT-PCR) .The capability of migration and invasion of LA-N -6 cells were evaluated by the transwell chamber assay.Results The expression of S100A4 and MMP-2 mRNA were decreased by 35.6 % and 25.5 % respectively compared to those in control group. The number of migrating and in vading LA-N-6 cells were significantly lower in the asODN group than those in the control group(P<0.05). Conclusions S100A4 asODN signifi cantly reduce the S100A4 mRNA expression and the migratory and invasive potenti al of neuroblastoma cells, with concomitant decrease of MMP-2 mRNA expression. Thus S100A4 gene may exert its effect on invasion and metastasis of neuroblastom a cells by stimulating the motility of tumor cells as well as influencing the ex pression of MMP-2 gene.
