摘要
目的 :探讨周期素激酶抑制剂p2 7在大黄素抑制肾系膜细胞 (mesangialcell,MC)增生中的作用。方法 :蛋白印迹 (west ern杂交 )方法测定MC裂解液p2 7蛋白水平 ,[3H]胸腺嘧啶核苷 ([3H]TdR)测定MC增生的情况 ,观察不同浓度大黄素 (50mg/L、10 0mg/L )对肿瘤坏死因子 α(tumornecrosisfactor α ,TNF α)刺激的MC中p2 7水平及增生程度的影响。结果 :TNF α (2 0万U/L)使无血清培养 2 4h的MC中p2 7水平降低 ,同时使 [3H]TdR掺入增加 ;大黄素提高TNF α刺激的MC中p2 7水平 ,同时使 [3H]TdR掺入减少。大黄素的上述作用随剂量的增大而增强。结论 :大黄素可能通过提高p2 7水平抑制TNF α诱导的MC增生。
Objective:To investigate the role of p27 in t he inhibition of emodin on the mesangial cell (MC) proliferation induced by tumo r necrosis factor-α(TNF-α). Methods: p27 protein of MC was detec ted with western blotting analysis. The degree of MC proliferation was estimated through [ 3 H]thymidine ([ 3 H]TdR) incorporation. Different dosa ge of emodin (50 mg/L,100 mg/L) was added into MC stimulated by TNF-α. R esults: TNF-α (200 kU/L) decreased p27 level of MC cultured in serum-fr ee DMEM for 24 hours and increased[ 3 H]TdR incorporation. Emodin increase d p27 level of MC stimulated by TNF-α and decreased [ 3 H]TdR incorporat ion. The more the emodin was added, the greater the above-mentioned effect of e modin. Conclusion: The increment of p27 level maybe play an importan t role in the inhibition of emodin on MC proliferation induced by TNF-α.
