摘要
目的建立龙胆中龙胆苦苷的RP HPLC定量分析方法 ,并对 3 8批不同产地的龙胆进行含量测定。方法采用DiamonsilC18色谱柱 ( 5 μm,2 0 0mm× 4 6mmi d ) ,以乙腈 水 醋酸 (V∶V∶V =1 0∶80∶1 )为流动相 ,流速为 1 0mL·min-1,紫外检测波长为 2 70nm ,柱温为室温 ,内标物为对羟基苯甲酸 ( 1 0 0mg·L-1)。结果龙胆苦苷在 1 2 8~ 1 5 3 6mg·L-1质量浓度内线性关系良好(r=0 9999) ,方法回收率为 99 1 % ,RSD为 0 4% (n =9)。 3 8批龙胆中龙胆苦苷的含量差异较大。结论该方法可用于质量控制 ,并为不同产地龙胆中龙胆苦苷的含量比较提供了依据。
Objective To establish a RP-HPLC method for the determination of the content of gentiopicroside in 38 samples of Radix Gentianae from different habitats.Methods A Diamonsil-C18 column(5 μm,200 mm×4.6 mm i.d.)was used with the mobile phase being acetonitrile-water-acetic acid (V∶V∶V=10∶80∶1),the flow rate being 1.0 mL·min-1.The detection wavelength was set at 270 nm,the column temperature was room temperature and the internal standard was p-hydroxybenzoic acid (100 mg·L-1).Results The calibration curve was linear (r=0.999 9)in the range of 12.8~153.6 mg·L-1 for gentiopicroside,the average recovery of the method was 99.1%,with RSD 0.4%(n=9).The contents in 38 samples of Radix Gentianae of gentiopicroside differ greatly.Conclusions This method can be used for the quality study of Radix Gentianae and for the contrast of contents of gentiopicroside from different habitats.
