香菇原生质体诱变及营养缺陷型突变体筛选 被引量:8

摘要 从香菇(Lentinus edodes)单核菌丝分离出的原生质体铺于含0.6mol/l蔗糖的CM表面,经紫外线照射30秒后,生长出的菌落被转移到MM之上。在MM上生长停止或生长缓慢的菌落经进一步检测,得到7个营养缺陷型突变体。经斜面CM上3次转代培养及原生质体分离和再生,表明有3个突变体仍保持稳定;可用作细胞融合研究,但其原生质体产量尚需进一步提高。 Protoplasts isolated from monokaryotic mycelia of Lentinus edodes were spread on CM containing 0.6 mol/1 sucrose and irradiated for 30 seconds. After colonies formed, they were transplanted on MM and those showing little growth were detected further. 7 auxotrophic mutants were obtained at last, among which 3 mutants remained stable after protoplast isolation, regeneration and mycelial subculture on CM slant for 3 times. They can be used in protoplast fusion in later studies, despite their protoplast yields were low and have to be improved.
