摘要
用HPLC紫外检测器研究了无紫外吸收化合物的负峰测试法 ,探讨了负峰测试法的原理。通过对乙酸酯系列的 5种样品的分析 ,确定了负峰测定的基本色谱条件 ,考察了乙酸酯系列测定的灵敏度、精密度、检出限和线性范围。该法为一些在紫外区无吸收或弱吸收物质的色谱分析提供了一种简捷、方便的手段和色谱机理研究的新方法。同常规的采用正峰测试样品的方法相比较 ,负峰法应用范围更宽 ,能使紫外检测器变为一种更通用的检测器。
The determination of low or non-absorption substances in ultraviolet region by reversed phase high performance liquid chromatography was carried out. By analysis of five homologues of acetic ester, the basic chromatographic conditions of negative-peak determination were established. The conditions including components of mobile phase, background substance, detection wavelength and flow rate were optimized. The precision, lowest detect-ability and the linear equation were also determined. The method is an efficient, simple and convenient way for high performance liquid chromatographic separation of non-absorption substance.
