摘要
目的 :探讨膀胱移行细胞癌核基质蛋白 (nuclearmatrixproteins ,NMPs)与膀胱移行细胞癌的相关性。方法 :根据Wen等的方法改良制备膀胱移行细胞癌NMPs ;NMPs免疫家兔获得抗血清 ,利用免疫吸附法及硫酸铵沉淀法纯化抗血清 ;Westernblot及confocal分析膀胱移行细胞癌NMPs与其他对照组之间的差异性。结果 :膀胱癌NMPs具有 (2 0~ 4 0 )× 10 3 及 (6 0~ 90 )× 10 3 两富集蛋白条带区域 ,与对照组比较差异明显 ;膀胱移行细胞癌中NMPs主要表达于细胞核、核膜及部分胞质 ,而对照组织无明显特异性表达 ,差异有显著意义。结论 :膀胱移行细胞癌存在肿瘤相关的特异性NMPs ,为开发基于尿液或血液的免疫检测方法提供了实验基础。
Objectives To explore the correlation between the differential expression of nuclear matrix proteins (NMPs) and the human bladder transitional cell carcinoma (hBTCC).Methods Materials were hBTCC fresh and related paraffin tissues,the hBTCC-NMPs were prepared from fresh tissues by using the Wen`s method with some modifications.Rabbits were immunized with the total hBTCC-NMPs.Antiserum was purified by immunoadsorption with antigen of mucous membrane of normal urinary bladder and sulfate ammonium precipitation.The hBTCC-associated NMPs was analyzed by Western blot and indirect immunofluorescence through confocal scanning microscope.Results The possible hBTCC-related NMPs of molecular weight (20-40)×10 3 and (60-90)×10 3 were found by Western blot analysis.Indirect immunofluorescence assay showed that the proteins of the NM were largely localized in nuclear and nuclear membrane in situ NM,and cytoplasm partly in hBTCC paraffin sections.Conclusions Carcinoma-related NMPs expresses in hBTCC.It may be a foundation for developing a new detection assay with urine or serum and suggesting a potential biomarker of hBTCC.
