摘要
目的 探讨普通重力和模拟微重力条件下构建人间充质干细胞的三维培养体系。方法 将人间充质干细胞和人发角蛋白共同置于普通重力和模拟微重力条件下进行培养。结果 当转速为 37.2r min时 ,人发角蛋白在旋转式细胞培养系统中呈悬浮状态 ,沿一条与氧交换膜的轴心垂直的环形轨道随液体流转动 ,1× 10 3个 ml的人间充质干细胞和人发角蛋白共培养 7d ,在模拟微重力条件下有较多的间充质干细胞贴附生长于人发角蛋白表面 ,密集、平行排列 ,细胞在人发表面几乎形成密集的一层 ,在普通重力条件下人发角蛋白表面生长的间充质干细胞较稀疏 ,大多呈多角形。结论 普通重力和模拟微重力条件均适合于间充质干细胞的三维培养 。
Objective To construct the human mesenchymal stem cells (MSCs) culture system for three-dimensional culture modeling under regular gravity and simulated microgravity.Methods The human MSCs and human hair keratin (HHK) were co-cultured under regular gravity and simulated microgravity.Results Human hair keratin was circumrotated in a near circular trajectory with the axis of oxygen-exchanged membrane in the fluid medium with a rotary speed of 37.2 rpm/min while avoiding collision with the wall of the rotary cells culture system.Human MSCs with 1×10 3/ml cell density adhered and grew well on the surface of HHK,with dense and parallel arrangment and almost formed a layer under simulated microgravity 7 days after culture.MSCs distributed less uniformly on the surface of HHK,and a majority of MSCs were in polygon shape under regular gravity.Conclusion Both conditions are suitable to construct human MSCs culture system for three-dimensional modeling,and the latter may be better.
