哮喘宁颗粒剂对哮喘大鼠糖皮质激素受体的影响

Effects of Xiao Chuan Ning Granules on Glucocorticoid Receptors in the Rats with Asthma

目的观察哮喘大鼠肺组织中糖皮质激素受体(GCR)、血清皮质醇的变化情况以及哮喘宁颗粒剂对其的影响,从而探讨调肝理肺法治疗哮喘的疗效机制。方法采用放射配基结合试验法测定各组大鼠肺组织中GCR的含量,用放射免疫法测定血清中内源性皮质醇的含量,将结果予以比较分析。结果哮喘模型组大鼠肺组织GCR最大结合容量和血清内源性皮质醇含量显著降低(P<0.05),经哮喘宁颗粒剂治疗后,GCR最大结合容量和内源性皮质醇含量均明显上调(P<0.05),与正常组无显著性差异。通过对照实验表明,哮喘宁颗粒剂对GCR和内源性皮质醇的良性调节作用与阳性对照药补肾防喘片相似。结论哮喘宁颗粒剂能够上调哮喘大鼠肺组织GCR和血清皮质醇水平,调肝理肺法治疗哮喘的疗效机理可能与此有关。

Objective Observing variation of Glucocorticoid receptors (GCR) in lung tissue and variation of cortisol in serum of rats with asthma and observing effects of Xiao Chuan Ning Granules on GCR and cortisol, so as to probe into mechanical of curative effect of Tiao Gan Li Fei on bronchial asthma. Method Detecting GCR maximum specific binding ability in asthmatic rats' lung tissues of different groups by the method of radio-ligand binding experiment, and content of cotisol in asthmatic rats' serum by the method of radio-immunity, then compare and analyse the result among different groups. Result Comparing with other groups, both GCR maximum specific binding ability and content of cotisol are lower in asthma rat group significently (P<0.05). After treat by Xiao Chuan Ning Granules, both GCR maximum specific binding ability and content of cotisol are up-ragulated compared with other groups (P<0.05). And the two markers have no significently different comparing with normal control group (P>0.05). By control experiment, it suggests that Xiao Chuan Ning Granules can benign regulate GCR and endogenetic cortisol similar as Bu Shen Fang Chuan Tablets (P>0.05). the latter is a kind of positive control medicion of asthma. Conclusion Xiao Chuan Ning Granules can up-regulated GCR maximum specific binding ability in asthmatic rats lung tissues and content of cotisol in asthmatic rats' serum. The mechanism maybe related with mechanical of curative effect of Tiao Gan Li Fei on bronchial asthma.
