摘要 目的 筛选亚甲蓝 (MB)光化学效应对血液中大肠杆菌的最佳灭活条件。方法 通过正交试验、单因素及双因素作用灭活细菌效果测定。筛选亚甲蓝光化学效应灭活血液中大肠杆菌的最佳条件及影响因素 ,观察其灭活效果。结果 正交实验显示 ,亚甲蓝光化学效应对血液中细菌灭活作用受 MB含量、卤钨灯照射强度和照射时间等因素的影响。试验筛选的最佳处理条件可以使大肠杆菌下降近 5个对数级。亚甲蓝与卤钨灯照射两因素的 T/ E值大于 1,具有协同增效作用。结论 亚甲蓝光化学效应灭活血液中大肠杆菌的最佳条件是 15μmol/ L亚甲蓝 ,4 0 0 0 0 L ux卤钨灯照射强度 ,4 0 min作用时间 ,0 .35 mmol/ Objective To explore the best inactivating condition of methylen blue (MB) for E.coli in the blood.Methods The best inactivating condition of MB for E.coli in the blood was screened by the orthogonal assay and the inactivating effect of single and double factors were determined.Results The results of orthogonal assays indicated that the inactivation efficacy for E.coli in the blood by MB photochemical treatment was influenced by MB content, the intensity and duration of halogen lamp irradiation. The best condition could reduce E.coli about 5 log. When the value of T/E of MB and irradiation of halogen lamp was bigger than 1, MB and irradiation of halogen lamp had synergetic effects.Conclusion The best inactivating condition of MB for E.coli in the blood was 15 μmol/L MB, 40000 Lux halogen lamp irradiation intensity, 40 min duration, and 0.35 mmol/l rutin.
