摘要
目的:通过成骨细胞与珍珠层/聚乳酸人工骨复合后在体内外增殖的研究,探讨珍珠层/聚乳酸人工骨的生物相容性。方法:将新西兰大白兔成骨细胞种植到珍珠层/聚乳酸人工骨材料上,用四唑盐(MTT)比色法、扫描电镜研究成骨细胞在体外增殖情况;在体外用5-溴脱氧尿嘧啶核苷(Brdu)标记成骨细胞,并将标记的同种异体成骨细胞接种于珍珠层/聚乳酸人工骨表面,复合培养1周后植入体内,采用免疫组化法,与体外检测孔对比,检测成骨细胞在体内的存活、增殖情况。结果:MTT法显示随培养时间的延长,细胞在材料表面不断增殖;电镜显示,体外细胞在材料表面及孔隙中附着、生长良好,存在接触抑制现象;成骨细胞与珍珠层/聚乳酸人工骨复合后能在体内继续存活并增殖。结论:珍珠层/聚乳酸人工骨对成骨细胞在体内外增殖无明显影响,具有良好的生物相容性。
AIM:To explore the biocompatibility of Nacre/PDLLA(N/P) artificial bone based on the study of proliferation of the osteoblasts loaded on Nacre/PLA artificial bone in vitro and in vivo. METHODS:The osteoblastic cells isolated from New Zealand rabbits were cultured and then seeded into the N/P artificial bone material in vitro.The proliferation of the osteoblasts was detected by MTT assay,observation of scanning electron microscope in vitro.The osteoblasts were labeled with thymidine analogue bromodeoxyuridine(Brdu) in vitro,and then implanted subcutaneously into N/P artificial bone.The survival and proliferation of the osteoblasts were analyzed by immunohistochemical staining at 1 week post implantation. RESULTS:The unceasing proliferation of osteoblasts was seen on the surface of material with MTT as time prolonged.Good adhesion,growth and inhibition of osteoblasts were seen on the surface and pore of the material in vitro.Osteoblasts could grow and proliferate in vivo after implantation into the N/P artificial bone material. CONCLUSION:N/P artificial bone has no obvious effects on the proliferation of osteoblasts in vitro and in vivo,it has good biocompatibility.
