摘要
目的 :探讨FHIT基因与乳腺癌发生发展的关系。方法 :运用PCR 变性聚丙烯酰胺凝胶电泳 银染法对FHIT基因位点上 5个微卫星位点进行LOH的检测。同时采用免疫组织化学方法检测了 40例肿瘤组织中Fhit蛋白的表达情况。结果 :3 7.5 %的原发性乳腺癌存在LOH ,45 %的肿瘤样本发生Fhit表达的缺失或明显下降 ,两者之间无显著性差异 (P =0 .0 0 0 )。结论 :FHIT基因位点的LOH及Fhit蛋白表达的缺失及下降是乳腺癌早期的和频发的事件 。
? Objective To explore the relationship between FHIT gene and the development and progression of breast cancer. Methods PCR denaturing polyacrylamide gel electrophoresis silver staining method was used to detect LOH of five microsatellite loci located in the FHIT gene site. Immunohistochemistry staining method was also used to show the expression status of Fhit protein in tumor samples. Results LOH was identified in 37.5% primary breast cancer. Lost or obviously decreased Fhit protein expression was showed in 45% cases. There was significant correlation between LOH of FHIT gene and loss of Fhit protein expression ( P =0.000). Conclusion LOH of FHIT gene and lost or decreased Fhit expression is an early and very frequent event in breast cancer. FHIT gene plays an important role in the development and progression of breast cancer.
