川芎嗪抗大鼠内毒素休克性肝损伤作用机理的研究

摘要 目的 :探讨川芎嗪抗大鼠内毒素休克性肝损伤的作用机理。方法 :将健康成年wistar大鼠 6 0只随机分为正常对照组、内毒素休克组、内毒素 +川芎嗪预处理组 ,以内毒素注射后 2h及内毒素休克组大鼠临终时为时点采集肝脏 ,采用western斑点印迹法及ELISA双抗体夹心法检测肝组织中HSP70及TNF -α含量。结果 :内毒素休克组大鼠较对照组肝组织HSP70及TNF -α含量显著升高 ,与川芎嗪预处理组大鼠相比 ,后者肝HSP70含量明显增加 ,TNF -α含量显著减少 ,动物存活时间延长 ,存活率显著提高 ;相关分析表明肝HSP70与TNF -α含量呈显著负相关。结论 :川芎嗪能显著提高内毒素休克大鼠肝脏HSP70含量 ,抑制肝TNF -α释放 ,并减轻内毒素诱导的肝损伤。 Objective:To probe into the effect of Tetramethylphyrazine(TMPZ)on HSP70 and TNF-α content in Liver tissue of rats with endotoxic shock.Methods:60 male adult rats were equally randomized to 3 groups:normal control,endotoxic shock and TMPZ groups.Western blot method and ELISA were applide to separately detect the hepatic HSP70 and TNF-α content 2 hours after endotoxin injection and the dying time of endotoxic shock rats.Results:Compared with normal control group,endotoxic shock troup saw a significant inerease of HSP70 and TNF-α content in Liver tissue,while TMPZ group,compared with endotoxic shock group,had a significant increase of HSP70 content,but a marked decrease of TNF-α content.Besides, the survival rate was highly enhanced in TMPZ group.Correlation analysis showed negative correlative relationship between HSP70 and TNF-α content.Conclusion:TMPZ can increase HSP70 content in Liver tissue of murine endotoxic shock, and suppress the release of TNF-α,so as to alleviate hepatic impairment induced by endotoxin.
