摘要
目的 探讨白细胞介素 - 18(IL - 18)基因转导对肺癌细胞系 PG细胞肿瘤原性的影响。方法 将携带人白介素 - 18的质粒 pc DNA3.1- h IL- 18转染到肺癌细胞系 PG中 ,通过药物新霉素 (G- 4 18Sulfate)进行筛选 ,利用RT- PCR、EL ISA方法对 IL- 18的表达水平进行检测。同时对裸鼠行皮下致瘤试验 ,观察 IL- 18转染前后 PG细胞致瘤性的改变。结果 IL- 18基因成功的转导入 PG细胞中 ,且能持续表达 ,转入 IL- 18基因的 PG细胞上清液 IL-18的生物学活性为 (187.5± 12 .3) U/ ml,接种裸鼠后无肿瘤长出。结论 PG细胞转入 IL- 18基因后将失去致瘤性 ,提示 IL - 18基因转导的
Objective To investigate the effect of interleukin 18(IL 18) gene transfer on the tumorigenesis of lung carcinoma cell line PG.Methods Human lung carcinoma cell line PG were transfered with lipofectamine and selected by G418.The biological activity of hIL 18 was tested by MTT method and nude mice were injected with PG cell with or without the hIL 18 gene.Results The hIL 18 cDNA was successfully integrated into PG cell.The biological activity was(187.5±12.3)U/ml.The nude mice injected with IL 18 gene engineered PG cell had no tumor growth.Conclusion The IL 18 gene engineered PG cell line may lost tumorigenicity.The PG cells transduced with IL 18 gene may be used as tumor vaccine.
