摘要 目的 制备rhBMP - 2聚氰基丙烯酸正丁酯纳米微球缓释系统 ,检测其对兔骨髓间充质干细胞(BMSCs)的生物学效应。方法 采用改良的乳化溶液聚合法制备rhBMP - 2纳米微球 ;采用MTT法检测微球对BMSCs的增殖状况的影响 ;碱性磷酸酶 (ALP)检测试剂盒检测细胞ALP活性以反映微球对其分化状况的影响 ,并与单纯rhBMP - 2的作用进行比较。结果 该微球缓释系统有生物活性 ,能显著促进BMSCs的增殖及分化 ,并呈剂量依赖性。其效应高于单纯施加rhBMP - 2的效应。 Objective To prepare the recombinant human bone morphogenetic-2 poly butylcyanoacrylate nanoparticles controlled release system, and to evaluate its biological effects on cultured rabbit bone mesenchymal stem cells.Methods The rhBMP-2 nanoparticles were prepared by improved emulsion polymerization method.Cell culture technique and MTT colorimetric assay were used to evaluate the proliferation of the cells,ALP kit was used to evaluate the ALP activity of the cells so as to show the differentiation of the cells by adding the rhBMP-2 nanoparticles or only rhBMP-2.Results Statistical analysis showed that rhBMP-2 nanoparticles had biological activity,and could enhance significantly both proliferation and differentiation of rabbit bone mesenchymal stem cells,The effect of the rhBMP-2 nanoparticles was significantly higher than that of rhBMP-2.Conclusion The effects of released growth factor controlled were significantly higher than common growth factor in tissue engineering work.
