CD_(44)V_6和MMP-9及P_(53)的表达与胃癌患者淋巴结转移的关系——附60例检测报告

Relationship between the expression of CD_(44)V_6, matrix metalloproteinase-9, P_(53) and lymphnode metastasis in gastric cancer.

目的:探讨CD44第6变异株(CD44V6)、金属基质蛋白酶-9(matrixmetalloproteinase-9,MMP-9)、P53蛋白表达与胃癌患者淋巴结转移的关系。方法:采用分子原位杂交和免疫组织化学方法对60例胃癌患者癌组织中的CD44V6信使核糖核酸(messengerribonucleicacid,mRNA)、MMP-9mRNA和P53蛋白的表达进行检测。结果:胃癌组织中CD44V6mRNA、MMP-9mRNA、P53蛋白表达阳性率分别为60%、63%、65%,CD44V6mRNA、MMP-9mRNA、P53蛋白阳性表达者的淋巴结转移率分别为92%、92%、90%,均明显高于阴性者的38%、32%、33%,P<0.05;同时有2个以上指标表达者的淋巴结转移率为100%,明显高于1个指标表达者(20%)和3个指标均不表达者(23%),差异均有统计学意义(P<0.05)。结论:CD44V6mRNA、MMP-9mRNA和P53蛋白的异常表达与胃癌淋巴结转移过程密切相关,对分析胃癌的生物学行为和判断预后有重要意义。

Objective: To evaluate the relationship between the expression of CD44V6, matrix metalloproteinase 9(MMP 9), P53 protein and lymphnode metastasis in gastric cancer. Methods: CD44V6mRNA, MMP 9mRNA and P53 protein were detected by in situ hybridization or immunohistochemistry in 60 gastric cancer tissues. Results: In gastric cancer tissues, the positive expression rate of CD44V6 mRNA, MMP 9 mRNA and P53 protein was 60% , 63% and 65% , respectively. In tissues with positive expression of CD44V6 mRNA, MMP 9 mRNA and P53 protein, lymphnode metastasis rate was 92% , 92% and 90% , respectively, significantly higher than tissues with negative expression(38% , 32% and 33% , P<0.05). The lymphnode metastasis rate of tissues with positive expression of two or more parameters was 100% , significantly higher than tissues with positive expression of only one parameter(20% ) or without expression of these parameters(23% ). Conclusions: The abnormal expression of CD44V6mRNA, MMP 9mRNA and P53 protein is closely associated with gastric cancer lymphnode metastasis and is important in analyzing biological behavior and predicting the prognosis of gastric cancer.
