摘要
比较了常规的CTAB法与改进的CTAB法对樟树总DNA提取的结果。在常规的CTAB法基础上,采取先破碎细胞收集细胞核,将存在于细胞质中的大量影响DNA提取质量的多酚等次生代谢物除去后,再加核裂解液释放DNA,经分离纯化,提得的DNA通过A260/A280比值的测定及电泳,限制性内切酶消化,PCR扩增,结果表明,用改进的CTAB法提取的DNA纯度和完整性均优于常规的CTAB法。
A comparative study on total DNA extraction from Cinnamomun Camphora which is rich secondary metabolites.Based on the conventional CTAB methods,an efficient procedure was developed.The major improvement included elimination a lot of secondary metabolites such as polyphenols etc,and collection of nuclei before free DNA.Then CTAB-free buffer was added to extract total DNA.The results from ratios of A_(260) and A_(280),electrophoresis,digestion with restriction endonucleases and PCR amplification suggested that the modified CTAB methods produce high quality DNA compared with conventional CTAB methods.
