芪箭消瘦汤对自身免疫性甲状腺炎小鼠TNF、IL-6的影响

Effects of Qijian Xiaoying Decoction on TNF, IL-6 of Autoimmune Thyroiditis in Mice

目的:观察芪箭消瘦汤对实验性自身免疫性甲状腺炎小鼠自身抗体、甲状腺病理、胀腔巨噬细胞诱生细胞因子的影响及作用机理。方法:运用人甲状腺球蛋白与完全弗氏佐剂混合免疫注射法模拟小鼠实验性自身免疫性甲状腺炎,用被动血凝法检测甲状腺自身抗体水平,收集腹腔巨噬细胞用放射免疫法测上清液肿瘤坏死因子和白细胞介素-6的含量,HE染色法观察小鼠早状腺组织病理改变。结果:芪箭消瘦汤明显抑制甲状腺自身抗体水平,减轻小鼠甲状腺组织淋巴细胞浸润。模型组小鼠腹腔巨嗜细胞诱生TNF、IL-6含量显著高于正常组(P<0.01)。芪箭小剂量组、雷公藤片组TNF显着低于模型组(P<0.01),大剂量组与模型组有差异(P<0.05)。芪箭大、小剂量组IL-6显著降低(P<0.01)。结论:芪箭消瘦汤可调节细胞因子的释放,改善桥本氏甲状腺炎的自身免疫紊乱,对小鼠实验性自身免疫性甲状腺炎疗效明显。

Observe the effects and mechanism of Qijian Xiaoying decoction to EAT in murine on autoantibodies, cytokines induced by mice peritoneal macrophage and pathology. Method: Injection of hTg and CFA can induce EAT. Survey autoantibody levels with PHA. Observe the pathology changes of thyroid tissue of murine with HE colouration. Gather murine peritoneal macrophage to investigate the levels of TNF and IL-6 with RIA. Results; Qijian Xiaoying decoction can suppress obviously autoantibody levels and alleviate lymphocyte soakage of murine thyroid tissue. TNF and IL-6 in model group were higher than that in control group (P <0. 01). The data of TNF in SQJ and TT group were markedly decreased ( P < 0. 01). The levels of IL-6 in BQJ and SQJ group were significantly lower than that of model group ( P < 0. 01 ). Conclusion : Qijian Xiaoying decoction can adjust autoimmune disorder and the releasing of cytokines in Hashimoto's Thyroiditis, which have distinctness curative effect on EAT.
