摘要 目的:探讨持续压力对体外培养的大鼠成骨细胞c—fos mRNA表达的影响。方法:取1—2d龄SD大鼠颅盖顶骨进行成骨细胞原代培养,采用压缩空气在密闭容器内对第2代细胞施以3×10^5Pa的静压力,分别于加力前、加力后1/2h、1h、2h提取细胞总RNA,用RT—PCR方法检测c—fos mRNA的表达。结果:大鼠成骨细胞在持续压力作用下,c—fos mRNA的表达呈一过性增强。c—fos mRNA的表达在受力1/2h后增强,受力1h后达到高峰,受力2h后恢复至受力前的水平。结论:c—fos可能参与了成骨细胞受到持续压力后机械信号转变为生物学信号的过程。 Objective:To investigate effects of continuous compression on the expression of c - fos mRNA in cultured rat osteoblasts in vitro. Method :Primary rat osteoblasts were obtained from parietal bones of 1 - 2 days old SD rats. The second generation cells were subjected to continuous compression of 3 × 105Pa by compressed air in a sealed container. Total RNA were extracted before application of compression(Oh) and 1/2 h,1 h,2 h after application of compression. The expression of c - fos mRNA were evaluated by reverse transcriptase - polymerase chain reaction( RT - PCR). Result: The expression of c- fos mRNA were increased transiently after application of compression in rat osteoblasts. The expression of c - fos was increased at 1/2 h after application of compression, peaked at Ih and recovered to the level of Oh at 2 h. Conclusion;c- fos maybe participate in the transduction of mechanical signal to biological signal when rat osteoblasts are subjected to continuous compression.
