碘化钠对人甲状腺上皮细胞分泌细胞因子IL-6和IL-8的影响 被引量:24

摘要 目的 研究碘化钠 (NaI)对人甲状腺上皮细胞 (TEC)分泌细胞因子IL 6和IL 8的影响 ,以探讨碘在Graves病 (GD)发病中的可能机制。方法 取手术切除的Graves病患者甲状腺组织和甲状腺腺瘤患者瘤旁正常甲状腺组织 ,进行细胞培养 ,以不同浓度的NaI(10 - 8~ 10 - 3mol L)刺激单层培养的甲状腺细胞 ,采用放射免疫测定技术测定刺激前、后甲状腺细胞培养上清液中细胞因子IL 6和IL 8的含量。同时以透射电镜观察NaI刺激后甲状腺细胞的形态学改变。结果 ①正常甲状腺细胞能分泌少量的IL 6和IL 8;GD的甲状腺细胞IL 6和IL 8的分泌量与正常TEC相比显著增加 (P <0 .0 1)。②当NaI浓度为10 - 8~ 10 - 3mol L时 ,正常人甲状腺细胞IL 6、IL 8的分泌量与NaI浓度 0mol L正常组相比无显著性差异 (P >0 .0 5 ) ;透射电镜显示正常甲状腺细胞无损伤型改变。③当NaI浓度为 10 - 6 ~ 10 - 3mol L时 ,GD甲状腺细胞IL 6的分泌量显著增加 (P <0 .0 5 ) ;当NaI浓度为 10 - 7~ 10 - 3mol L时 ,GD甲状腺细胞IL 8的分泌量显著增加 (P <0 .0 5 )。NaI浓度超过 10 - 6 mol L时 ,透射电镜显示GD甲状腺细胞出现损伤型改变。结论 高浓度的碘不仅造成GD甲状腺细胞的直接损伤 ,而且可以增加甲状腺细胞细胞因子IL 6和IL 8的分泌 ,特别是在GD? Objective To investigate the influence of sodium iodide(NaI) on cytokines (IL-6,IL-8) secretion of human thyrocytes and to study the effect of iodine on the pathogenesis of autoimmune thyroid disease. Methods The human thyroid epithelium cells(TEC) from Graves' disease(GD) patients and para-adenoma normal tissues from thyroid adenoma patients were cultured in the absence or presence of 10 -8-10 -3 mol/L NaI.The contents of cytokines such as IL-6,IL-8 in supernatant of cultured thyrocytes were detected by radioimmune assay (RIA). Results ①A few IL-6,IL-8 were detected in supernatant of normal thyrocytes. IL-6,IL-8 in supernatant of GD thyrocytes increased significantly compared with normal thyrocytes (P<0.01). ② When NaI concentration was 10 -8-10 -3 mol/L, the contents of IL-6,IL-8 in supernatant of normal thyrocytes was not significantly different from that of 0 mol/L NaI group (P>0.05);TEM showed that normal thyrocytes did not have damaging change. ③ When NaI concentration was 10 -6-10 -3mol/L, the contents of IL-6 in supernatant of GD thyrocytes were significantly higher than that in 0 mol/L NaI group (P<0.05). When NaI concentration was 10 -7-10 -3 mol/L the contents of IL-8 in supernatant of GD thyrocytes were significantly higher than in 0 mol/L NaI group (P<0.05).When NaI concentration was over 10 -6 mol/L, TEM showed that GD thyrocytes showed damaging changes. Conclusion High concentration of iodine not only results in direct damage of GD thyrocytes,but also increases cytokines (IL-6,IL-8) secretion of normal and GD thyrocytes, especially in GD thyrocytes. High concentration of iodine plays a very important role in the pathogenesis of GD.
