摘要
人参寡糖素M能提高红花(Carthamus tinctorius)培养细胞的生长速率和培养细胞中代谢产物α-生育酚的含量。其最适作用浓度在愈伤组织培养中为5mg/L。在红花细胞悬浮培养加入人参寡糖素M1d后,培养细胞中α-生育酚的含量即提高,但由于累积效应,因而于细胞接种当天同时加入人参寡糖素M对细胞生长和α-生育酚含量的提高效果较好。加入人参寡糖素M可缩短红花悬浮培养细胞生长的延缓期,并于指数生长期作用最明显;另外可使细胞生长及α-生育酚积累同时提前达到最高值,因而缩短了细胞收获时间。
Oligosaccharins are oligosaccharides from cell walls which have regulatory functions on plant morphogenesis and secondary metabolism, and can elicit the formation of phytoalexins. The physiological effects of one of the oligosaccharins on cultured cells of Carthamus tinctorius were studied. The oligosaccharin used was isolated from cell cultures of Panax ginseng and was given the name of ginseng-oligosaccharin M. It was found to promote the growth of and α-tocopherol formation in cultured cells of C. tinctorius(Table 1). Its optimum concentration on callus culture was 5 mg/L (Fig. 1). From a study of the effects of addition of ginseng-oligosaccharin M on different days from the starting of cell growth, it was seen that its effect on cell growth and α-tocopherol formation was evident the second day of its addition any time during the culture up to 20 days, but the total effect was the largest when it was added on the first day, because of the cumulative effect (Table 2). When ginseng-oligosaccharin M was added to a culture of suspended cells, the duration of exponential growth was shortened and the cells could be harvested earlier, because of the promotive effect on growth. The cell growth rate during exponential growth stage was increased by 21%~23% and the α-tocopherol content was increased by 1. 4- to 1. 8-fold by the addition of ginsengoligosaccharin M(Fig. 2).
