硫丹对PC_(12)细胞增殖的影响及诱导其凋亡的研究 被引量:4

摘要 目的 :观察硫丹 (ES)对培养的PC12 细胞增殖和形态学的影响及其诱导PC12 细胞凋亡作用。方法 :不同浓度MPP+ 和ES分别干预PC12 细胞 2 4h ,利用TUNEL染色、流式细胞仪、电镜检测观察其结果。结果 :① 1mmol·L-1MPP+ 组细胞数明显下降并有形态学改变 ;而 0 0 3mmol·L-1ES组细胞数就明显下降和有形态学改变 ,以 0 0 1mmol·L-1组最明显。②MPP+ 和ES均可诱导PC12 细胞凋亡 ,电镜下 0 0 6mmol·L-1ES组有较多凋亡细胞和少量坏死细胞。结论 :ES对PC12 细胞有毒性作用 。 Aim:To investigate the effect on the proliferation and morphology of the cultured PC 12 cells,and the induction of apoptosis on PC 12 cells by endosulfan.Methods:The various concentrations of MPP + and endosulfan were added to the cultured PC 12 cells for 24 h,the proliferation and morphology of the cells and the induction of apoptosis ratio of the cells by endosulfan and MPP + were observed using the TUNEL staining,FCM,EM technology.Results:① PC 12 cells were significantly decreased after exposure to 1 mmol·L -1MPP + and the morphology alse changed.With 0.03 mmol·L -1 endosulfan,cells were significantly decreased and the morphology of the cells were changed,the most obvious chang of PC 12 cell was in 0.1 mmol·L -1 endousulfan ②Both MPP + and endosulfan can induce PC 12 cells apoptosis.Under EM,Many of apoptotic cells and a few of necrotic cells can be identified in 0.06 mmol·L -1 endosulfan.Conclusion:Endosulfan had toxic effects on PC 12 cell.The possible mechanism may be related to the apoptosis of the cells.
