摘要
目的 研究活化的基质金属蛋白酶-2(aMMP2)及其组织抑制因子(TIMPs)在皮肤损伤和修复过程中的动态平衡变化。方法 建立以STZ诱导的糖尿病大鼠深Ⅱ度烫伤模型,测定烫伤后0、1、3、7、21 d组织培养上清液中aMMP-2和TIMP-2含量。结果 烫伤后糖尿病和正常鼠皮肤组织释放的aMMP-2和TIMP-2逐渐升高。糖尿病组未损伤组织释放的aMMP-2明显高于正常组(P<0.05),伤后21d TIMP-2明显低于对照组(P<0.05);正常组aMMP-2/TIMP-2比值基本稳定于一定的水平,而糖尿病组aMMP-2/TIMP-2比值逐渐下降,自伤后7 d起,显著低于0时相点(P<0.01)。结论 糖尿病者刨伤前后皮肤组织中基质金属蛋白酶及其相关的生物活性物质的平衡均出现异常。
Objective To investigate the change of aMMP-2 and TIMP-2 in the wounds of diabetic and non-diabetic burned mice. Methods The streptozotocin( STZ) induced diabetic SD rats, inflicted with 10% tissue burned surface area(TBSA) of deep partial thickness scalding , were adopted as the model. The concentrations of aMMP-2 and TIMP-2 were observed by analyzing the fluid samples of cultured tissue on postburn day(PBD) 0, 1, 3, 7, 21. Results Compared with normal rats, aMMP2 was much higher in the uninjured diabetic skin tissue, TIMP2 was obviously lowered on PBD 21. After burn injury the ratio of aMMP2 to TIMP2 was in a steady status in the normal group but gradually decreased in the diabetic group. Conclusion The balance between MMPs and their inhibitors is abnormally in diabetic skin tissue before and after injury.
