摘要
目的 :观察冠心病 (CHD)患者外周血内皮祖细胞 (endothelialprogenitorcells,EPC)数量和功能的改变。方法 :选择CHD患者和非CHD患者各 2 0例 ,用密度梯度离心法从外周血获取单个核细胞 ,将其接种在人纤维连接蛋白包被培养板 ,培养 7d后贴壁细胞进行细胞化学分析。激光共聚焦显微镜鉴定FITC标记荆豆凝血素Ⅰ和DiI 标记的乙酰化低密度脂蛋白双染色阳性细胞为正在分化的EPC。采用二苯基四氮唑嗅盐比色法、改良的Boyden小室和黏附能力测定实验观察EPC的增殖能力、迁移能力和黏附能力。 结果 :CHD患者外周血EPC数量明显减少 [(31.8± 7.7)∶(5 9.5± 10 .6 )EPC/视野 (× 2 0 0 ) ,P <0 .0 5 ],且CHD患者外周血EPC的黏附能力、迁移能力和增殖能力也明显受损。结论 :CHD患者外周血EPC数量减少、功能减退。
Objective:To investigate changes in the number and activity of endothelial progenitor cells(EPC) from peripheral blood in patients with coronary artery diseases.Method:EPCs were isolated from patients with coronary heart disease(CHD,n=20) and age-matched control subjects (n=20). EPC were characterized as adherent cells double positive for DiL-acLDL-uptake and lectin binding by direct fluorescent staining under a laser scanning confocal microscope. EPC proliferation and migration were assayed by MTT assay and modified Boyden chamber assay, respectively. EPC adhesion assay was performed by replating those on fibronectin-coated dishes, then adherent cells were counted.Result:The number of EPC was significantly reduced in subjects with CAD compared with that of age-matched control subjects ( 31.8± 7.7)∶( 59.5± 10.6)EPC/×200 field; P< 0.05). In ad- dition, the functional activity of EPCs such as proliferative, migratory and adhesive capacity was also impaired in patients with CAD.Conclusion:It is suggested that a novel pathophysiological mechanism of CAD may be defined: the reduction of EPCs with decreased functional activity.
