摘要
目的:检测9-顺-维甲酸处理前后人肺腺癌细胞株PG、A_(549)、SPC-A_1的cyclinD1、cdk4转录水平的变化,探讨9-顺-维甲酸抑制肺腺癌细胞生长的分子机制。方法:采用相对定量逆转录PCR方法,分析细胞周期因子cyclinD1、cdk4转录的变化。结果:9-顺-维甲酸既可以降低PG、A_(549)的cyclinD1转录(P<0.01),又可以降低PG、SPC-A_1的cdk4的转录(P<0.01或P<0.05)。结论:9-顺-维甲酸对肺腺癌细胞的生长抑制作用,与其调控细胞周期因子cyclinD1、cdk4的表达密切相关。
AIM:To investigate the mechanism of 9-cis-RA inhibiting the growth of lung adenocarcinoma cells, and we detect the expressional changes of cyclinD1 and cdk4 in lung adenocarcinoma cells PG, A_(549), SPC-A_1 before and after being treated with 9-cis-retinoic acid(9-cis-RA). METHODS: RT-PCR was used to analyse the transcriptional changes of cyclinD1 and cdk4 in PG, A_(549) and SPC-A_1. RESULTS:9-cis-RA decreased the transcription of cyclinD1 in PG and A_(549)(P<0.01), and also decreased the transcription of cdk4 in PG and SPC-A_1(P<0.01 or P<0.05) at the dosage of 1×10^(-6) mol/L. CONCLUSION: The growth inhibitory effect of 9-cis-RA on lung adenocarcinoma cells may be tightly correlated with its mediating the transcription of cyclinD1 and cdk4.
