摘要
研究了新试剂对羧基苯基荧光酮(p CPF)与钼的显色反应,建立了一种测定微生物细胞质中微量钼的光度法。在硫酸和阳离子表面活性剂CTMAB介质存在下,p CPF与Mo(Ⅵ)发生灵敏的显色反应生成红色配合物,其最大吸收峰位于528.5nm,表观摩尔吸光系数为1.12×105L·mol-1·cm-1,Mo(Ⅵ)含量在0~2.0μg·ml-1范围内符合比耳定律;室温下显色反应迅速完成,而钼配合物吸光度可稳定12h以上。此外,该方法还具有良好的选择性,常见离子及大量蛋白、核酸、糖等生物活性物质均不干扰钼的测定,已用于杆菌和酵母细胞质中钼的测定,结果满意。
In a dilute sulfuric acid medium and in the presence of CTMAB, Mo(Ⅵ) reacts with p-CPF to form a red complex having its absorption maxima at 528.5nm and an apparent molar absorptivity of 1.12×10~5L·mol^(-1)·cm^(-1). Beer′s law holds in the range of 0~2.0μg·ml^(-1) of Mo(Ⅵ). The complex forms instantaneously and keeps stable for at least 12h. Those cations and anions commonly encountered and large quantities of protein, nucleic acid sugars were found to have no interference with the reaction. This method has been used with good precision and accuracy in the determination of trace amounts of molybdenum(Ⅵ) in cytoplasm fluid.
