摘要
目的研究半胱天冬酶鄄3(caspase鄄3)在异丙基肾上腺素(isoprenaline,Iso)致大鼠急性心肌损伤中的作用及碱性成纤维细胞生长因子(bFGF)对它的影响。方法将Wistar雄性大白鼠随机分成3组:对照组、损伤组和bFGF保护组,光镜观察心肌组织的病理变化,TUNEL法检测心肌细胞凋亡,免疫组化和原位杂交方法检测Caspase鄄3蛋白及mRNA的表达水平,底物降解法测定该酶的活性。结果Iso损伤组心肌坏死较重,有显著心肌细胞凋亡,且Caspase鄄3基因mRNA及蛋白表达水平明显升高,酶活性增加;bFGF保护组心肌坏死明显减轻,凋亡细胞数减少,Caspase鄄3基因mRNA和蛋白表达水平明显下降,酶活性受到抑制。结论bFGF可能通过抑制Caspase鄄3的表达和活性减少心肌细胞凋亡而减轻大鼠心肌损伤。
Objective To study the role of Caspase-3 on myocardial injuy induced by isoprenaline (Iso) and the effects of basic fibroblast growth factor (bFGF) on Caspase-3. Methods Wistar rats were divided into three groups at random. They were the control group, the Iso group and the bFGF group, respectively. The pathological changes were observed by microscope. The apoptosis were observed by TUNEL. The mRNA and protein expression of Caspase-3 were tested with immunohistochemistry and in situ hybridization methods. Results In the Iso group, the myocardial necrosis were more serious, and there were more apoptosis. Moreover, the expression levels of mRNA and protein of Caspase-3 increased, and the enzyme activity of Caspase-3 went up. In the bFGF group, the myocardial necrosis were relieved and there were less apoptosis. The expression levels of mRNA and protein of Caspase-3 decreased clearly. Condusion The bFGF can alleviate rat myocardium from injury by inhibiting the expression and activity of Caspase-3 and reducing myocardial apoptosis.
